Is it possible to specify a minimum quantity for a product when the wholesale module is activated in an account? For example, 5 apples, otherwise it becomes a retail sale anyway?

I‘m sorry, there is no direct option to do it.
But a possible workaround would be to setup an individual product with zero discount for wholesalers and a 5 item bundle product with wholesaler discount.
It would also be possible with quantity options/attributes.
You may just download the Addon and check out all options.
